1.  Why should I switch to using this medium?

Extensive validation is conducted on every lot of our AB2™ Basal Neural Medium and ANS™ Neural Medium Supplement to eliminate batch-to-batch variability and to provide continuous quality product to the end user.  Prequalified AB2™ Basal Neural Medium and ANS™ Neural Medium Supplement provide consistent results and ArunA Biomedical’s complete medium formulation is specifically engineered for the optimal health and propagation of neural cells.

2.  How do you qualify or validate your medium and supplement?

Each lot of AB2™ Basal Neural Medium and the ANS™ Neural Medium Supplement is extensively validated.  Each lot undergoes the standard sterility, pH, osmolarity and endotoxin testing.  Most importantly, unlike other commercially available media, each lot of AB2™ Basal Neural Medium and the ANS™ Neural Supplement is evaluated for growth performance on live cultures of neural progenitors.  For each lot, the growth of live cultures is assessed over 10 passages for proliferating neural progenitors and over 2 weeks differentiation into post-mitotic neuronal cells.  Any lot affecting growth behavior or karyotype is discarded.  Thus end-users are assured that batch-to-batch variability is eliminated using the AB2™ Basal Neural Medium and the ANS™ Neural Supplement.

3.  What cell lines or types can I use this complete medium with?

Since the AB2™ Basal Neural Medium and the ANS™ Neural Medium Supplement are specifically formulated for neural cells, ideally these products can be used for the culture of neural stem cells and progenitors derived from human embryonic stem cells (hESCs) and induced pluripotent stem cells (iPSCs).  These products can also be used with their differentiated derivatives (mature neuronal and glial phenotypes).  These products are compatible with cells grown on Matrigel, laminin, gelatin, collagen, poly-L-lysine, poly-O-ornithine and any other commonly used substrates to culture neural cell types. 
 

4.  Does this medium contain protein, animal components, serum, albumins?  Does this medium and supplement qualify for clinical applications?

The AB2™ Basal Neural Medium and the ANS™ Neural Medium Supplement together provide a serum-free, complete culture medium for neural cells.  The AB2™ Basal Neural Medium is free of any animal and protein components.  However, the ANS™ Neural Medium Supplement does contain bovine serum albumin (BSA).
Together as a complete medium, the AB2™ Basal Neural Medium and the ANS™
Neural Medium Supplement do not qualify as xeno-free and are not suitable for clinical applications. 

5.  Is this medium pre-made or do I need to mix components?

To make the complete medium, two 5 mL vials of ANS™ Neural Medium Supplement are thawed and added to 500 mL of AB2™ Basal Neural Medium, then sterile filtered.
ANS™
Neural Medium Supplement is stored at –20°C and AB2™ Basal Neural Medium is kept at 4°C.  Once thawed, the ANS™ Neural Medium Supplement should not be re-frozen (avoid freeze/thaw cycles).  Both the AB2™ Basal Neural Medium and ANS™ Neural Medium Supplement are guaranteed for one year post manufacture if stored properly. 

6.  Are there any required but not supplied materials needed for use with cells?

For the culture of proliferative human neural stem cells or progenitors, especially if you are using ArunA Biomedical’s hNP1™ Neural Progenitor line, the addition of basic fibroblast growth factor (bFGF or FGF2) and L-glutamine to the AB2™ Basal Neural Medium and the ANS™ Neural Medium Supplement is required, while leukemia inhibitory factor (LIF) and antibiotics are optional.  We recommend the following brands: recombinant human LIF (Millipore | Cat# LIF 1010), bFGF (R&D Systems | Cat# 233-FB).

7.  Can I add other supplements to this medium?

The AB2™ Basal Neural Medium and the ANS™ Neural Medium Supplement are formulated to provide versatility to the end user.  Not only can this medium be used as a growth medium for the culture of proliferative neural cells, but it makes an exceptional base medium for the addition of specific growth factors or other proteins for directed differentiation to specific neural phenotypes (i.e. dopaminergic neurons, motorneurons, astrocytes, etc.).

Tips for using Matrigel:

  • When you receive the Matrigel stock, immediately move to 4°C to thaw.
  • Once thawed, mix with equal volume of DMEM media while keeping all materials on ice (make 1:2 solution).
  • If desired, aliquot solution into chilled tubes and freeze at -20°C.
  • Only use thawed Matrigel stock solution for 2 weeks post thaw.
  • When making 1:200 solution, place all tubes, bottles on ice.
  • Only use 1:200 Matrigel solution 2 weeks post preparation.
  • If desired, rinse the tissue culture plate with PBS ++ prior to Matrigel coating.
  • When coating, keep the Matrigel on ice and be sure to add enough solution to cover completely the surface of the plate.
  • Immediately move the plate to the 4ºC for 1-3 hours OR leave the coated plate in the hood at room temp for 1-3 hours; avoid keeping plates in the fridge overnight, this is more likely to make them dry out.
  • Observe plates under the scope prior to use to look for an unusual amount of debris/cracks/dry patches.
  • Rinse with PBS++ prior to plating with cells.
